Additional Information: | Aqueous Two Phase extraction for the extraction and purification of Horseradish Peroxidase from Radish was carried out. Different PEG/Salt systems were tried at different %TLL for the better recovery of the enzyme Horseradish Peroxidase. At the optimized conditions ATPE comprising of PEG 6000/Potassium Phosphate at a %TLL of 19.91 with a Volume Ratio of 0.15 resulted the Specific Activity of 9094.91(U/mg) with a yield of 90.80%. From the overall results a purification factor of 9.88 was obtained for the enzyme Horseradish Peroxidase. The purified enzyme was then subjected to Ultrafiltration for the removal of phase components and then the purified enzyme was dried using three different drying methods that are Freeze drying, Vacuum Shelf drying and Spray Drying. It was done with the help of encapsulating agents like Maltodextrin, Polymer and ATPS as such. Out of the all drying methods Freeze Drying was found to be the best resulting in a Specific Activity of 308.26(U/mg) with ATPS used for encapsulation. It was also found from the investigation work that encapsulation by ATPS showed better results in terms of specific activity of enzyme than the other encapsulating agents in all the three drying operations. |
