Additional Information: In the present investigation, polyphenol oxidase and peroxidase activities in whole wheat flour, refined wheat flour, bran and germ were determined and these enzyme activities were found to be the highest in germ. As very little data is available on the biochemical properties of polyphenol oxidase from wheat, polyphenol oxidase from germ was partially purified using ammonium sulphate precipitation followed by gel filtration and obtained a 10 fold purified enzyme. The germ polyphenol oxidase showed wide substrate specificity compared to that of isolated from endosperm. The germ polyphenol oxidase exists different isoforms and they can be broadly classified into two groups, low molecular weight and high molecular weight isozymes. The germ polyphenol oxidase showed less activity below pH 8 and temperatures above 40C.
