Abstract

This Dissertation / Report is the outcome of investigation carried out by the creator(s) / author(s) at the department/division of Central Food Technological Research Institute (CFTRI), Mysore mentioned below in this page.

Item Type: Student Project Report
Additional Information: One of the high growth areas in fermentation technology is the search for new bioactive molecules from microbial sources since natural products are an excellent source of chemical structures with a wide variety of biological activities. Besides antibiotics, some of the bioactive molecules include mammalian enzyme inhibitors, mycotoxins, herbicides, insecticides etc. One such group of bioactive metabolites produced by fermentation is low molecular weight enzyme inhibitors. Tyrosinase is a multifunctional, glycosylated, and copper-containing oxidase, which catalyzes the first two steps in mammalian melanogenesis and is responsible for enzymatic browning reactions in damaged fruits during post-harvest handling and processing. Neither hyperpigmentation in human skin nor enzymatic browning in fruits are desirable. These phenomena have encouraged to seek new potent tyrosinase inhibitors for use in foods and cosmetics. A trial has been made to screen and produce an inhibitor of Tyrosinase from microbial source. The present work deals with the screening and selection of microbial cultures for the effective production of Tyrosinase inhibitors. Isolation of these cultures and fermentation is carried. After the primary screening of fungal isolates, the potent inhibitor is purified by thin layer and column chromatography with different solvent ratios. The results suggest that the inhibitor has an IC 50 value of 10.5 μg/ml. The purity of the crude and purified compound is determined by HPLC. Further studies are required to determine the structural characteristics and mode of inhibition of the inhibitor.
