Additional Information: In view of the growing concern of population to cure diseases by natural means and greener technology the present study was undertaken. Five different cultures were screened for protein, biomass content, and BSH activity. The culture with best BSH activity was selected and was used for probiotic food preparation. The culture strain was improved for maximum BSH activity by adapting it to different bile salts. The adapted culture strains were screened for cholesterol assimilation. This improved strain which could reduce cholesterol to a higher level was used in the preparation of a probiotic cholesterol lowering high nutritive millet based blend product. Studies were conducted on the product for a period of one week for shelf storage at room temperature of 30 C which could reduce cholesterol at a higher level and the quality could be kept along with higher culture viability. Sensory evaluation was done on the product by a trained panel at the sensory Science department of CFTRI, Mysore, India. The product received very good acceptability with a score of 7.5 on hedonic scale of ten. A novel rapid assay method using HPLC was standardized for the estimation of cholesterol during the study which has been reported for the first time in probiotic millet based blend product and is not known in the literature so far. The important minerals such as sodium and calcium were determined by atomic absorption spectroscopy and the levels increased by a half fold for sodium and for calcium an increase of one sixth in the probiotic millet based blend over the control sample. These minerals may help to increase cell metabolism and osteoporosis.
