Abstract

Cancer is a complex devastrating disease with increase incidences in recent years. Despite the availability of many anticancer drugs. Disease pathogenecity profile convincingly evidenced that this is due to metastatic spread for which successful therapy is not available . In this direction we have identified from the laboratory that targeting galectin-3 would be a potential therapeutic approaches in addition to the use of antioxidants, cytoprotective and DNA protective compounds since celluar DNA damage mediated by oxidative radicals are the soul cause of the disease. For this the current study on the 2-hydroxy 4-methoxy benzaldehyde (HMBA) and its modified derivative Oximes is succesful in many bioactivity assays performed. The free radical scavenging activity of the derivatives 2-hydroxy 4-methoxy benzaldoxime (HO) and 2,4- di hydroxy benzaldoxime (DHO) showed 5 and 30 folds increase in activity. Besides in other bioactivities such as cytoprotectity, H+K+ATPase inhibitory activity, Tyrosinase Inhibitory activity, DNA protectivity indicated that either no significant differences or HMBA showed much better than the derivatives. Since the current study had yielded a better antioxidant-sister molecule combination of HMBA and its derivatives may work choesively and arrest many of disease advancing reactions. Also the bioavailibilty studies indicated substantially good results unlike other phenolics and could be confirmed after tissue distribution studies. Besides, a natural carbohydrate polymers from Bael (Aegle marmelos) showed good galectin-3 mediated metastasis inhibition in in vitro experiments. Since no treatment is available for this, the natural polymer could be used to develop strategic anticancer compoun from natural source. The galectin-3 inhibition was further substatiated with amount of galactose (19%) and high amount of Uronic acid. In whole the study showed potential multi step role of HMBA, HO, DHO and Pectic from bael as depicted in scheme1 could be used as potential therapy in progression of diseases like cancer and ulcer.
