Additional Information: | <p align="justify">The aim of this study was to understand different algorithms and mathematics used in various phylogenetic methodologies of tree building analysis.The second aim of this study was to understand and evaluate the workings of the various methods used in the assessment of phylogeny and to identify the method that gives the correct tree based on the sequences under study. Most of the work hasbeen carried out using the software – Molecular Evolution and Genetic Analysis. A number of sequences for the EF1 gene from different Fusaria were compared using different methods. These comparisons were also made on the EF1 sequences representing only by the exon region. Finally the protein coded for the EF1 sequences were also used to build trees. The EF1 gene from four unidentified clones of Indian isolates were amplified by PCR and cloned for the purpose of conducting molecular phylogeny analysis. The clones were sequenced at DNA Sequence Lab, Delhi University, Delhi. Sequences were then compared and aligned with ClustalX with random Fusarium sequences of EF1 from the GenBank database using various different tree building methods and using different models of substitutions available from MEGA2. Trees were compared and a consensus about the Fusarium species had been derived as being not highly divergent when compared with unknown sequences and the choice of suitable method for the sequences under study was identified.</p> |
