Additional Information: | In response to problems associated with plastic waste and its effect on the environment, there has been considerable interest in the development and production of biodegradable plastics. Biopolymers is used to refer to those polymers formed in nature during the growth cycles of all organisms. They are also referred to as natural polymers. Polyhydroxyalkanoates (PHAs) are polyesters that accumulate as inclusions in a wide variety of bacteria. PHAs are regarded as an attractive source of non-polluting plastics and elastomers that can be used for speciality and commodity products. Biopolymers include polysaccharides, starch, cellulose from plants and microbial carbohydrates like Xanthan, dextran, gellan and pullalan produced by fungi. Polyhydroxy butyrate (PHB) is the best known member of the PHA series of polyesters. With this background the following objectives i.e. Isolation of PHA from soil, characterization, production, extraction, purification and structure analysis by FTIR, NMR was studied. Also the PhaB, Pha C genes which codes for AcetoacetylcoA reductase and synthase enzymes were studied. The organism was observed to produce PHA and identified as gram positive rods. The morphological and taxonomical evidence shows the organism is found to the genera. Amphibacillus based on Bergey’s Manual of Determinative Bacteriology (9th Edition). It is found suitable to grow utilizing glucose as a sole carbon sources at pH 7.2 at 30oC at 200 rpm. The organism is also found to contain both Pha B and Pha C enzyme which are important in PHA biosynthetic pathway and is a potent producer of polyhydroxyalkanoates. |
