Abstract

Culltures of Haematococcus pluvialis were exposed to mutagens, such as UV and EMS (ethyl methanesulphonate), in order to isolate mutants with enhanced astaxanthin productivities. Results showed that the survival rate decreased with increasing UV exposure time and increasing EMS concn. Mutants were screened further using inhibitors of the carotenoid biosynthetic pathway, viz. diphenylamine (15-90muM), nicotine (160-320muM) and compactin (1.5-3.0M). The mutants thus obtained showed enhanced (2.2-3.2-fold) astaxanthin accumulation and also exhibited higher lycopene cyclase activity.
