Abstract

The effect of a supplement of DL-ethionine on tissue acid mucopolysaccharides (MPS) of rats maintained on a low-protein diet (9% casein) was studied. The incorporation of 35S (Na35SO4) into lipids and MPS was also studied. MPS were isolated from tissues after proteolytic digestion by pepsin and trypsin. Liver intestine and kidney MPS showed increased amounts, in g of tissue, of uronic acid and hexosamine in the ethionine-fed group. Increases in associated protein and neutral sugar content were observed in intestinal and liver MPS. Fractionation of liver and intestinal MPS showed increases in the "chondroitin sulphates" in the ethionine-fed group. Intestinal MPS isolated after digestion of tissue by pronase showed decreased MPS content (uronic acid) and rduced incorporation of 35S in the ethionine-fed group. This decrease was prevented by methionine supplementation. The studies showed that ethionine administration results primarily in an accumulation of neutral glycoproteins and alters the proportion of sulphated and non-sulphated tissue MPS.
