Abstract

This Dissertation / Report is the outcome of investigation carried out by the creator(s) / author(s) at the department/division of Central Food Technological Research Institute (CFTRI), Mysore mentioned below in this page.

Item Type: Student Project Report
Additional Information: Indian poultry is producing nearly 180,000 tons of viscera which is rich in proteins & proteolytic enzymes. Proteases that catalyze the cleavage of the peptide bonds in Proteins(beg et al., 2003)comprise a large group of enzymes that are ubiquitous in nature. Proteases are mainly derived from animal, plant and microbial sourses. These enzymes are important in a number of diverse and crucial biological processes; for example, they are involved in the regulation of metabolism And gene expression, by employing aqueous two phase extraction comprising of PEG-sodium phosphate &potassium phosphate purification of acidic proteases is efficiently carried out. The study indicated the ATPS system consisting of PEG 8000 with sodium phosphate and potassium phosphate can be effectively used for separation and purification of acidic proteases from chicken intestine. However, as multiple factors influence the behavior of protein and enzymes in an aqueous-two phase system, it is necessary to evaluate the combined effect of all the factors involved. Once the major factors influencing the system are identified, it is desirable to optimize the conditions necessary for the effective separation and purification of alkaline protease from chicken intestine using ATPS and to conduct scale-up studies.
