Additional Information: | Production of thermoactive chitinases by local isolates of Aspergillus flavus, Fusarium oxysporum and Penicillium monoverticillium were carried out under solid state fermentation using commercial wheat bran. A. falvus produced maximum 26.37 unit/ g of initial dry substrate (U/g IDS) of endo-chitinase and 222.1 U/g IDS of β-Nacetyl- D-hexosaminidase. F. oxysporum produced 13.28 U/g IDS of endo-chitinase and 168.3 U/g IDS β-N-acetyl-D-hexosaminidase. P. monoverticillium produced 41.0 U/g IDS of endo-chitinase and 195.4 U/g IDS β-N-acetyl-D-hexosaminidase. The crude enzyme showed maximum activity at 62±2oC and pH 4.6 for endochitinase. The chitinases obtained from Aspergillus flavus, Fusarium oxysporum and Penicillium monoverticillium were concentrated by dialysis using high molecular weight polyethylene glycol. The concentrated chitinase systems were effective in hydrolysis of colloidal chitin and production of N-acetyl-D-glucosamine, indicating its potential for industrial applications. |
