Additional Information: The existing energy crisis has instigated the search for alternative sources which include the bio-fuel production from the edible crops like soyabeans, corn potatoes etc., as well as the algal species like Botryococcus braunii and Chlorella vulgaris which have the capacity to accumulate the lipid in their tissues. The lipid extracted can be treated with ethanol and can be used as bio-fuel by mixing it with the conventional petrol. This study examines ways to magnify the lipid yield from alga. In this study, the algal species chosen was Scenedesmus, known for its high biomass yield. The growth of the alga was monitored by providing the nutrients like nitrogen and phosphorous in three different ratios to find out in which ratio of nitrogen and phosphorous, growth was better. Growth was also estimated by the determination of the chlorophyll content present. To harvest the biomass of the grown alga, flocculation with various flocculants was carried out. The biomass harvested was dried using the lyophilization technique. The lyophilized biomass was then used for extraction of lipid. The calorific value of the biomass was determined using the Bomb Calorimeter. As the cell wall of alga was thick, the study also carried out experiments to disrupt the cell wall of the alga which included crushing, refluxing, steam extraction and sonication, of which steam extraction gave good results. The extracted lipids were then transesterified to separate the fatty acids from the other constituents like carbohydrates and nucleic acids. The trans-esterified samples were then analyzed using Gas Chromatograph and the lipid profiles were compared.
