Abstract
β‐glucosidases (EC 3.2.1.21) play an essential role in efficient hydrolysis of cellulosic biomasses. By hydrolysis of cellobiose, β‐glucosidases relieve inhibiting conditions, and enhances hydrolysis of cellulose bycellobiohydrolases and endoglucanases.The feasibility of using coffee husk for the production of β-glucosidase by Rhizopusstoloniferinsolid-state fermentation was evaluated in this study.The effect of pH (3-7), moisture (40-80%) and fermentation duration (72-168 h) on β- glucosidaseproduction was investigated using statistical optimization method. A three variable five level central composite design was used for the study. The partial purification was carried out using activated charcoal and ammonium sulphate (80%) and the enzyme was characterized to determine the optimum pH, temperature and the effect of different metal ions and inhibitors on the enzyme activity.The fermentation variables on optimization revealed maximum β-glucosidase production atpH 5.2, moisture 53%with fermentation duration of 112 h. The enzyme was purified 1.7 fold using ammonium sulphate.It had optimum parameters of 60°C temperature, pH 6.0, Km59 mM, and Vmax 78 μmol mg−1min−1 with cellobiose as a substrate. The enzyme activity was furtherenhanced by 1.15 and 1.2 fold in presence of Ca2+ and Mg2+ metal ions. The activity was inhibited in the presence of SDS, Fe3+ and Zn2+. These results indicated that coffee husk as a sole carbon sourceprovided necessary nutrients for cell growth and also β-glucosidasesynthesis. To the best of our knowledge, this is the first report on β-glucosidaseproduction utilizing coffee husk.The present study demonstrates that Rhizopusstolonifercanefficiently be used to achieveβ-glucosidase production with an economical and environmentalbenefit in solid-state fermentation.
