Additional Information: In the present investigation, kinetics of transfructosylation reaction by FTase from Aspergillus oryzae MTCC 5154 was studied. Crude enzyme was used for the production of FOS at lab, bench and pilot scale and hence, crude enzyme has been used for the determination of kinetic parameters. The enzyme was produced by submerged fermentation and the crude enzyme was used for the kinetic studies. Kinetic parameters (kcat, Km and Ki) were determined from the experimental data at various substrate concentrations and also in the presence of glucose. Transfructosylation was inhibited by glucose by mixed inhibition (Ki d-glucose = 0.8M). Time course of FOS indicated that percentage of FOS had reached a maximum of 28% by the eighth hour of incubation.
