Abstract

This Dissertation / Report is the outcome of investigation carried out by the creator(s) / author(s) at the department/division of Central Food Technological Research Institute (CFTRI), Mysore mentioned below in this page.

Item Type: Student Project Report
Additional Information:
Biodegradable plastics are the ones that can be broken down into simpler substances by the activities of living organisms. A variety of truly biodegradable polymers like polylactic acid (PLA), thermoplastic starch and poly-hydroxybutyrate/valerate are available, of which poly-hydroxybutyrate (PHB) and Polyhydroxyalkanoates (PHA) have gained more importance. The PHA production studies done with the A. latus MTCC 2309 indicates that the organism was able to utilize lactose as a carbon source however the yield was negligible.Glucose was found to be an ideal carbon source for maximum production of biomass and PHA. Soil extract broth medium was found to be the best medium for primary inoculum preparation. The results of the intracellular β-Galactosidase activity of A. latus MTCC 2309 suggests that the organism was capable of producing the enzyme β-Gal and its activity was maximum in the case of lactose. The sucrose hydrolase activity was found to be the maximum in case of Mauha flower extract than the other carbon sources. The PHA production studies carried out with the Mauha flowers suggests that it could be used as a cheaper carbon source for the PHA production. PCR amplification of the phaC shows evidence for the presence of CAB operon. The results of the GC and FTIR conclude that A. latus MTCC 2309 is capable of producing copolymer (PHB-Co-PHV) in juice extract and hence this strain can be exploited for the industrial production of PHA co-polymer.
