Additional Information: |
Extensive review of research reports published in the area of membrane processing of enzymes revealed that the ultrafiltration membranes were best suited for concentration and purification of enzymes. The molecular weight of enzyme ranges from 10-100 kDa and the researchers generally used a 10 kDa UF membrane independent of the molecular weight of the enzyme. However, a proper selection of membrane process specific for each process is desirable. In this investigation, a set UF membranes varying in MWCO (10-100 kDa) were screened considering the reported the molecular weight of polygalacturonase (~42 kDa) produced by the mutant strain of Aspergillus carbonarius. A MF (150 nm) membrane was used as a prefilter for reducing the polysaccharide content in the culture filtrate. This fungi also produces xanthophylls (pigment) having an average molecular weight of 574 Da. The pigment is extracted using organic solvents and partial desolventization was attempted using a hydrophilic NF membrane.
|
