Additional Information: Induction of cellular injury / cell death by Lindane;role of oxidative stress with specific reference to ROS; and amelioration of cellular oxidative stress by antioxidants was investigated. Mechanism of lindane induced cytotoxicity was studied in EAT cells. Lindane caused dose and time dependent cytotoxic injury to EAT cells as evident from cell viability and LDH leakage. Lindane induced the generation of ROS in a dose-dependent manner and depleted GSH level which led to oxidative stress and ultimately cell death. Induction of DT-diaphorase was a characteristic feature of lindane cytotoxicity to EAT cells. Quercetin, a bioflavonoid ameliorated the cytotoxic effect of lindane by suppressing ROS generation, glutathione depletion and cell death. The novel antioxidant, 4-hydroxyisophthalic acid from D.hamiltonii showed cytoprotective effect against lindane cytotoxicity at a much lower concentration, suggesting its ameliorative potential.
