Additional Information: E.faecium is considered as an important organism for dairy industry. In dairy industry, b-galactosidase enzyme is rather used as crude extract to prevent high cost of purification. Thus it is important to characterize the enzyme in crude extract of b-galactosidase producers.The influence of different medium on growth and b-Gal production by Ent.faecium was studied. After identifying the lactose fermenting capacity of strain Enterococcus faecium MTCC 5153 it was decided to explore lactose-based medium for the production of b-galactosidase. Three media were used by substituting the carbon source for the production of b-galactosidase. It was found that maximum cell biomass was obtained in MRS broth. In another experimental trial, low-cost TGE was tested. It was found that glucose do not induce b-galactosidase production so negligible activity was observed. Glucose of TGE was substituted with lactose and TYE was formulated which showed a very high titre of b-galactosidase production. Based on the above results and the utilization of lactose as a carbon source, production of b-galactosidase in whey permeate (WP) was studied. Normal cell growth and a high titre of b-galactosidase were obtained but less than TYE. In our experiment to facilitate healthy cell growth, 6ml of WP was supplemented with 2% YE. Ent. faecium showed maximum activity when grown in the presence of 2% lactose, followed by skim milk. But no b-galactosidase activity appeared when it is grown in the presence of glucose.
