Abstract
Lipase was isolated from defatted rice bran by a new procedure which yields higher enzyme activity and quantity of enzyme. The procedure involves selective ammoniuum sulphate precipitation followed by ion exchange chromatography using DEAE-Sepharose CL-6B gel and column chromatography. The purified and homogeneous enzyme was characterized for optical, physicochemical and hydrodynamic properties. The enzyme had a UV absorption max. at 276 nm with an extinction coeff. value of 15.25. Fluorescence excitation and emission max. were 285 and 344 nm, respectively. Specific activity was 18 units, and the enzyme had a pH optimum of 7.5 and temp. optimum of 30
