Abstract

Groups of weanling rats were maintained on a diet deficient in vitamin A; control rats received a vitamin A supplement. Rats at the acute stage of deprivation, when no liver stores of vitamin A were detectable, were given injections of acetate-1-14C and palmitic acid-1-14C and were killed after 24 h. Vitamin A deprivation resulted in a decrease in triglycerides and a relative increase in free fatty acids of intestinal lipids. Incorporation of acetate-1-14C was the same in triglycerides, whereas it was higher in other lipid constituents. In contrast, palmitic acid-1-14C was incorporated to a greater extent in all but the cholesterol ester component in the deprived group. Significant 14C activity from palmitic acid was associated with the free cholesterol fraction in the deprived group whereas no incorporation was observed in the intestinal cholesterol of normal rats. It is suggested that, in vitamin A deficiency, the oxidation of higher fatty acids and the utilization of the "acetate" derived from palmitate for synthesis of cholesterol are enhanced.
