Additional Information: Fumonisins are mycotoxins which are secondary metabolites and synthesized by certain filamentous fungi. These are usually produced on contaminated corn and corn derived products. These are produced under certain favourable environmental conditions such as proper moisture, sufficient oxygen, suitable temperature, physical damage of the commodity etc. Fumonisins have been found to health hazards to human and animals. These toxins are estimated by TLC and HPLC techniques which need sophiscated equipment, extensive cleanup etc. Hence, immunological methods were optimized for on- field detection of fumonisin in food and feed samples. Antifumonisin antibodies were obtained by injecting toxin-protein conjugate to poultry. The antibodies IgY were isolated and purified by PEG method. The protein profile indicated that IgY of 7th week old egg yolk had highest protein. However, Ig Y of 3rd week old egg yolk gave highest antibody titre. An antigen and antibody dilution of 1:50,000 and 1:2.5 lakh respectively, were found to be optimum by checker board analysis. Fumonisin level of 1pg could be detected by Dot-ELISA. The signal amplification using biotin and avidin combinations showed good amplifications of the visual colour. The method developed was used to estimate fumonisin levels in market sample.
