Additional Information: | The production of polyhydroxyalkanoates form bacteria has gained interest since they are biodegradable and ecofriendly. These bioplastics are better suitable for conventional polymer of petroleum origin. But the production cost of this biopolymer is much expensive then the convectional polymer produced from petroleum origin. Hence there is a need to reduce production cost by using cheaper sources. In the present study, optimization of medium for PHA and amylase was carried out by using inexpensive carbon sources (potato starch, corn starch, soluble starch etc). Potato starch has shown to produce good yield of PHA. Effect of nitrogen sources on PHA and amylase was also studied. Nutrient supplements like rice bran hydrolysate, wheat bran hydrolysate individually and in combination were used for production of PHA. Response surface methodology (RSM) was used to optimize fermentation medium for Bacillus cereus. For PHA production by RSM, rice bran, ammonium acetate, potato starch, Na2HPO4 and KH2PO4 were considered for the optimization of medium components. Different combinations of medium (33) were designed by using these variables and fermentation was carried out. Based on the results obtained it was tested with biostatical analysis by using Quattro pro software. Optimization of media by conventional method was also carried out with different parameters (temp, pH, rpm and inoculum percentage). It was concluded that rice bran which is rich in protein, starch etc enhanced the production of biomass as well as PHA and appear to good supplementation. Potato starch can be used as a cheaper carbon sources for Bacillus cereus. RSM study can be effectively used for the production of PHA by Bacillus cereus. The use of rice bran hydrolyzed reduces the cost of production. Further work should be carried for the characterization of the polymer by GC, NMR and FTIR analysis. |
