Additional Information: | In the present study, twenty five different fungi have been isolated from the soil samples and screened for their capability to produce alpha-glucosidase inhibitor. Each fungal culture was fermented in submerged (PDB) and solid state fermentation (wheat bran) for the crude production. The fermented broth was extracted in ethyl acetate, and vaccum distilled (solvent evaporation). The crude compound were redissolved in DMSO and A process for isolation of α-glucosidase inhibitor was established. From the secondary screening, Aspergillus niger strain was found to be the effective producer of the α-glucosidase inhibitor. Therefore, the selected culture was grown on large scale and the crude ethyl acetate extract was purified to homogeneity (89% TLC pure). The purified compound was little viscous and had an Rf value of 0.34 on TLC plate and had λmax of 340.8nm. Further studies like Preparative HPLC, GCMS , NMR, etc. would help determine the chemical structure of the active molecule. Further work has to be carried out to purify the inhibitor in order to enable the structural elucidation. Further studies like optimization of growth conditions, strain improvement techniques, etc. will certainly help in the better production of the inhibitor. |
