Additional Information: The main aim of the investigation “Screening of Topoisomerase II inhibitor for health application” was to study the topo. II inhibitors from fungal culture isolated from soil. One of the most important problems of modern medicine is the fight against several non-infectious and infectious diseases. Cancer is one among the primary causes of mortality in almost all countries. Screening of anti tumor compounds uses several types of test systems. Among them bacterial test system is common and efficient for primary screening. The interest in bacterial models is determined not only by their convenience for work or inexpensiveness, but also because the microorganism and a tumor cell have several common characteristics. They may be exemplified by mutants with an oxidation deficiency resembling oxidation in a tumor cell. One of them, E. coli DH5-α is the closest to a tumor cell and turned out to be most adapted to selection of producers of antitumor antibiotics, which is more sensitive than other microorganism. In the course of screening of topoisomerase II inhibitors, fungal strain from the soil sample collected from various regions was found to produce topoisomerase II inhibitor. Ethyl acetate extract of the fungi after week fermentation, unidentified fungus (U3, U8, U16 and U18) was found to produce topoisomerase II inhibitors. Among these U8 was more effective against E. coli DH5α and less toxic as it showed less inhibition against E. coli wild type. Further purification by column chromatography, TLC and HPLC showed the compound was partially purified. One of the fraction from column chromatography showing good inhibition was taken for further study. Dose dependent antibacterial assay was performed, which showed extracted inhibitor was effective at dose 40μg/10μl. It was also standardized with Doxorubicin®. Inhibition period was also tested by bacterial growth curve. The inhibitor started to show its effect from 1 hour and which was good at 8 hours. It effectively shortened the log phase of bacterial growth. So, the inhibitor screened showed good result up to this but, still there is need of further purification and further studies to test its effectiveness.
