Additional Information: | Background: The Plants Zingiber officinale (Zingibereace) and Brassica oleracea (Brassicaceae) have been used traditionally as anti-ulcer dietary sources. Literature also suggests the positive role of phenol bound polysaccharides in inhibiting gastric ulcers. The current study was carried out to substantiate data in this regard. Objective: The present study was focused on the evaluation of pectic polysaccharide extract Zingiber officinale rhizome and Brassica oleracea leaves, and their pharmaceutical formulations for ulcer preventive and ulcer healing efficacy in various experimental animal models. Method: The antiulcer efficacy of GRPP, CBPP and their respective tablet and syrup formulations were evaluated in pyloric ligation induced ulcer model, swim-stress induced ulcer model and indomethacin induced ulcer model. Ulcer index, H+K+-ATPase inhibition, gastric mucin, oxidative stress markers and histopathology studies were done to evaluate the anti-ulcer efficacy. To substantiate the results, an approach was made to determine the possible mechanism by various in-vitro assays including H+K+-ATPase inhibition; free radical scavenging effect and reducing power assay; and cytoprotectivity and DNA protectivity against oxidative damage. The bioavailability of formulations of the extracts was also evaluated. Result: In different animal models, GRPP/CBPP and their formulations found to produce significant antiulcer activity with decrease in ulcer index and H+K+-ATPase activity (p< 0.001, p<0.01). Enhancement in mucoprotection and oxidative stress regulations was also noticed. Upon in-vitro estimation GRPP and CBPP were found to have H+-K+- ATPase inhibition, muco-protective and anti-oxidant activities with variable potency. In the entire study, GRPP was found to be comparatively better than that of CBPP. The syrup formulation of GRPP gave highly significant potency among all in all in-vivo models. Conclusion: The results are suggestive of anti-ulcerogenic potency of GRPP, CBPP and their formulations at 50 mg/kg b.w. They are found to act by multiple mechanisms, mainly H+-K+-ATPase inhibition, partial anti-secretory, mucoprotective, cytoprotective and oxidative stress regulating mechanisms. |
