Abstract
Metastasis, inflammation and uncontrolled melanogenesis triggered by cancer cells via galectin-3, cytokines and dysregulated tyrosinase have been implicated in aggressive skin cancer condition. Cyclophosphamide and kojic acid although are used as immunomodulator and tyrosinase inhibitor respectively, the side effects posed by them warranted the need for safe alternatives. In the current study, we identified a, ∼91 kDa xylorhamnoar- abinogalactan I pectic polysaccharide from Bael (Aegle marmelos L.) - BAPP1 with constituting sugars - →4)β-D- Xylp-(1 → 3)-D-GalpA-(1→ xylogalacturonan and →4)α-D-GalpA-(1–2)-α-L-Rhap-(1→ rhamnogalacturonan structure with →5)α-L-Araf-(1→;→3)α-L-Araf-(1→ arabinan and →1)β-D-Galp-(4→ galactan as side chains. BAPP1 could potentially inhibit galectin-3 at MIC of 2.98 µg/mL; immunoregulatory cytokines IL10/IL17 (∼3 folds) and tyrosinase (IC50 of 28.0 ± 3.61 µg/mL), which are the key targets for the control of skin cancer (Scheme 1). Results were substantiated by both in vitro and UV/DMBA induced skin cancer models together with gene analysis. Xylorhamnoarabinogalactan I showed improved gut microbiota status in experimental animals. BAPP1 thus may find potential against metastasis.
