Enhancement of secondary metabolite production in hairy root cultures of Beta vulgaris and Tagetes patula under the influence of microalgal elicitors.

Abstract

Effects of aqueous extracts of the green alga Haematococcus pluvialis and the blue-green alga (cyanobacteria) Spirulina platensis on accumulation of betalaines (food colorant) and thiophenes (insecticides) in hairy root cultures of Beta vulgaris (red beet) and Tagetes patula (marigold), respectively, were studied. Upon treatment with the extracts (100 mug dry cell extract/ml of culture medium) of H. pluvialis and S. platensis, there was an increase in biomass of cultured B. vulgaris hairy roots; values were 165.3 g fresh wt./l (14.38 g dry wt./l) and 149.4 g fresh wt./l (13 g dry wt./l), respectively, and the initial inoculum was 1.25 g fresh wt./l (0.115 g dry wt./l). Accumulation of betalaines increased 2.28-fold on the 15th day in hairy root cultures treated with H. pluvialis compared with the control (7.9 mg/g dry wt.), from the initial level of 0.67 mg/g dry wt. Similarly, in S. platensis extract-treated hairy roots, accumulation of betalaines increased 1.16-fold on the 25th day compared with the control (11.08 mg/g dry wt.), from the initial level of 0.67 mg/g dry wt. It is concluded that S. platensis extract influenced accumulation of betalaines in B. vulgaris hairy roots, whereas H. pluvialis extract augmented betalaine and thiophene production in hairy root cultures of B. vulgaris and T. patula, respectively.
