Additional Information: This project is aimed at the production of the enzyme lipase from Aspergillus niger. On the basis of earlier experiments, Aspergillus niger was selected for the production of the enzyme lipase. This project encompasses the production of the enzyme lipase secreted extracellularly by the organism Aspergillus niger and to standardize the cultivation parameters in the shake flask experiments; followed by scaling up of the process using a bioreactor. This project is an attempt towards developing a process for the production of the enzyme lipase. Economical nutrient sources were used in an attempt to not to increase the media cost. The aim of this project was to scale up of enzyme and application of crude lipase for degreasing of leather; as enzymatic degreasing was found to combat pollution problems caused by the use of solvents and surfactants. The main objective of the present work was to observe production of the enzyme lipase using filamentous fungi Aspergillus niger in shake flask conditions and scaling up the process in a fermentor batch. Seven shake flask experiments and one fermentor trial was carried out. The maximum lipase activity obtained was 14 Units/ml at 48th hour. Total sugar, Reducing sugar and Protein of the samples were also determined.
