Additional Information: In the present study, bacterial lipases are reported .To use any lipase for industrial application, it is important to characterize the enzyme and study its properties. Therefore, in this study, screening, production and characterization of lipase from psychrophilic bacterial strains has been reported. The strains were identified by their growth characteristics on different substrates when the type of carbon source and the initial pH of the culture medium were optimized. The bacterial strains were screened for lipase production using the Rhodamine plate assay and four lipase producing bacterial strains were selected. The lipase production was measured with varying pH, temperature and various metal ions. Statistical analysis indicated that the variation in lipase production was significant between the bacterial strains. DNA from these bacterial strains was extracted and the isolates were subjected to gene amplification by PCR using specific primers. One of the amplified PCR product was selected for cloning of lipase gene and subsequently the plasmid isolation was done.
