Additional Information: | Fungal soft rot pathogens like C. paradoxa once after successful infection resume growth inside the host. To grow inside it has to obtain nourishment from the host. In case of C. paradoxa the successful colonization depends on the pathogen penetration of the host cell wall by using extra-cellular hydrolytic enzymes. The phenomenon of pathogenic growth is studied in this work and broth culture growth dynamics and extraction of chemical marker entities and estimating them under standardized procedures were presented. In case of liquid broth culture the estimation of fungal biomass is straight forward, filtration of fungal biomass and weighing. In case of chemical estimation the chitin has to be hydrolyzed to monomeric glucosamine and with the help of standard calibration graph it has to be estimated. By using internal standards the percentage recovery of the chemical also should be calculated. Ergosterol is extracted and measured us calibration curve and percent recovery needs to be calculated under the standard procedure In this investigation the developing biomass, sugar substrate consumption, production of proteins and the activity of extracellular cellulase during the pathogenesis are explained. The interrelationship of the individual parameter in the disease development is highly correlated. As this is a host-pathogenic relationship the understanding of control of obtainment of nourishment and development of biomass. |
