Additional Information: Antibiotics are invaluable drugs in the management of infectious disease. Unfortunately, their value is being jeopardized by the presence of microorganisms resistant to their effects. Since then, a succession of new antibiotic has been required to cope with the ongoing emergence of new strains of resistant bacteria, which have rendered even newly formed drugs ineffective. The competition between new drug development and the emergence of new resistant organism continues today. Currently, the most serious concess is that the bacteria are winning the race. Hence most of the researchers attention has turned towards the probiotic LAB cultures, which produces the antimicrobial substances. Cholera is one of the major water borne diseases, which emerge out as an epidemic that too in developing countries where the knowledge about the hygienic practice is much low so, we concentrate on the partial purification and characterization of the antimicrobial substance against V. cholerae A total of 5 LAB cultures were screened for their antagonistic activity against V. cholerae, of these LMU was selected and the strain was improved by chemical mutagenesis method for the maximum antagonistic activity against V.cholerae. The optimum condition for the growth of LMU was studied and the antimicrobial substance production was maximum al 30Oc at ph 5.5 in the stationary phase. The partially purified antimicrobial substances was prepared by acetone-methanol precipitation method and the purification fold was increased by 5 times whose molecular mass corresponds to the result of SDS-PAGE. The antimicrobial substance was proteinaceous in nature and heat stable because its activity was not inactivated by treatment with proteolytic enzyme and heat. Also the antimicrobial substance was compared with the antimicrobial substance was compared with the market antibiotic. Though the antibiotic showed maximum antagonistic activity against V. cholerae, the side effects,which were caused by them, were many. Such a kind of risk was not associated with the antimicrobial substance produced by the probiotic LAB culture.
