Additional Information: | Lipases(Triacyl glycerol hydrolases) catalyze the hydrolysis of ester linkages in lipids with the release of fatty acids and alcohol. Lipases can be used in leather processing for degreasing of skin/hide. Use of enzyme for degreasing gives better quality leather and have environmental advantage. On the basis of a number of experiments done earlier, Aspergillus niger MTCC 2594 was selected for the production of extracellular Lipase. The scope of this project is standardisation of parameters to produce lipase on larger scale using bioreactor and downstream processing. The objective of this work is to produce crude lipase for leather processing. Optimization of fermentation process parameters (i.e. pH, Dissolved oxygen, Temperature, Batch time) and downstream processing are worked out in this project work. A number of shake flask experiments have been conducted by varying the parameters. The maximum Lipase activity obtained was 20.0 U/ml at 100 rpm in a rotary shaker. The results at shake flask level indicated that the aeration does not effect the production of lipase. A number of in fermentation experiments in 2L and 5L volume fermentors have been conducted. The results obtained in the fermentor experiments have been reasonably comparable to those in the shake flasks. For downstream processing,filtration, microfiltration and ultrafitration using 10KDa membrane is suggested and the purification of lipase was found to be 5 fold. Further fermentor experiments and downstream processing studies will be needed for scale up and process standardization. |
