Abstract

The relationship between spot concn. and the number of components separated during micro-TLC of BHC was investigated. Graded concn. (0.025 to greater than 1.5 mug) of BHC were spotted onto micro-TLC plates, developed with n-hexane and sprayed with chromogenic reagent o-tolidine. 2 components (alpha- and gamma-BHC) were observed with a spot concn. of 0.025 mug BHC; 5 components (heptachlorocyclohexane and alpha-, beta-, gamma- and delta-BHC) were observed at spot concn. greater than0.25% BHC. Comparative trials with macro-TLC (BHC concn. 1 to greater than 40 mug) showed micro-TLC to be 40 times more sensitive. For semiquantitative determination of BHC, it is desirable to dilute the insecticide extract to yield 2 spots with either micro- or macro-TLC as greater than 2 spots represents a wide range in spot concn. and increases the error in determination of the insecticide. The technique may be used for determination of BHC residues in grain or other foods.
